IL-6 receptor / gp130 signaling via the JAK/STAT pathway
IL6R, JAK1, JAK2, TYK2
IL-6 Responsive Luciferase Reporter HEK293 Cell Line
– assay format: 96-well format
– construct: firefly luciferase reporter gene under the control of STAT response elements, stably integrated in HEK293 cells (BPS Bioscience)
– stimulus: recombinant human IL-6
– readout: IL-6-induced, JAK/STAT-dependent luciferase activity (luminescence)
– reference inhibitors: Tofacitinib, Ruxolitinib
– characterize JAK-targeting compounds
– support SAR and lead optimization
– validate mechanism of action in a cellular setting
– reduce development risk through early functional profiling

IL-6 Responsive Luciferase Reporter HEK293 cells were stimulated with increasing concentrations of recombinant human IL-6. Luciferase activity is shown as fold induction over unstimulated cells (EC50 = 1.6 ng/ml).

Inhibition of IL-6-induced reporter activity by the reference JAK inhibitor Tofacitinib (CP-690,550). Cells were treated with increasing concentrations of Tofacitinib and stimulated with 10 ng/ml human IL-6. Data are shown as percent luminescence relative to stimulated control (IC50 = 0.022 µM).
