THP-1 human acute monocytic leukemia cell line from ATCC

96-well plate format using Caspase-Glo 1 Inflammasome Assay

THP‑1 cells were primed with PMA for 3 days. The cells were then treated with the reference compound MCC950 (starting at 10 µM, 10-dose with 3‑fold serial dilutions) and subsequently stimulated with LPS for 3 hours. Caspase‑1 activity was measured using the Caspase‑Glo 1 Inflammasome Assay on an EnVision 2104 Multilabel Reader. IC50 value was calculated and IC50 curve was plotted using the GraphPad Prism program based on a sigmoidal dose response equation.
